If you're looking for a robust alternative to fluorescent and/or luminescent reporter mRNAs, look no further than Messenger Bio's NGFR mRNA. We engineered this mRNA to encode a truncated version of the human nerve growth factor receptor, such that it expresses only the cell surface and transmembrane domains of NGFR but lacks the cytoplasmic domain responsible for downstream cellular signaling. In this way, NGFR acts as a true functional reporter without adversely affecting normal cellular function. Our NGFR mRNA produces robust and easily detectable expression in different cell types, and is a viable alternative to other reporters that fail to fully recapitulate the translation and processing mechanisms required for expression of complex transmembrane proteins.
Messenger Bio’s NGFR mRNA can be used in functional reporter assays without requiring a cell permeabilization step, thus streamlining your experimental workflow. Simply transfect your cells with our NGFR mRNA and then assay protein expression via flow cytometry and/or confocal imaging, using any commercially available antibody directed against the surface domain of human NGFR.
Our NGFR mRNA has been further engineered for robust expression by inclusion of a Cap1 structure, highly expressing 5’ and 3’ UTRs, a 100-nt poly(A) tail, and a codon-optimized open reading frame. Choose from unmodified or modified bases (e.g., N1-methylpseudouridine, 5-methoxyuridine).
Our robust purification process removes dsRNA contaminants and other reaction components, generating highly pure mRNA that is capable of inducing very high levels of protein expression in diverse cell types, while causing minimal cellular innate immune activation (virtually undetectable).
As with all our mRNAs, this NGFR mRNA comes ready-to-use in a 1 mM sodium citrate buffer (pH 6.5), which is ideal for preserving RNA stability and extending shelf-life. Just complex with your favorite transfection reagent and you're ready to go. Alternatively, speak to us about our ready-to-transfect LNP formulations.
