Messenger Bio’s CRISPRa mRNA enables gene specific transcriptional activation—use this together with your guide RNA (gRNA) of interest to unlock increased expression of a target gene. Our CRISPRa mRNA encodes a CRISPR Cas9 effector without endonuclease activity (i.e., a dead Cas9; dCas9) fused to a transcriptional activator domain. When co-transfected with a well-designed complementary gRNA, this CRISPRa complex is directed to a specific promoter site in the genome, where it recruits important co-factors and RNA polymerase to initiate transcription of the downstream gene(s).

Our CRISPRa mRNA incorporates a Cap1 structure (biologically superior to Cap0), a highly expressing 5’ UTR, a highly stable 3’ UTR, a 100-nt poly(A) tail, and a codon-optimized open reading frame. Choose from unmodified or modified (N1-methylpseudouridine or 5-methoxyuridine) bases. Both types of modified bases have been shown to enhance protein expression in vitro and in vivo.

Our robust purification process eliminates dsRNA contaminants and other reaction components, resulting in highly pure mRNA capable of inducing very high protein expression levels in diverse cell types, while causing almost no innate immune activation.

Like all our mRNA, our CRISPRa mRNA comes in a ready-to-use 1 mM sodium citrate buffer (pH 6.5). This buffer ensures optimal RNA stability and shelf-life. Simply complex with your preferred transfection reagent and you’re ready to go. Alternatively, speak to us about our ready-to-transfect mRNA-LNP formulations.
